Glutamine is the most abundant amino acid in the body. It is made naturally within the muscles and other tissues for use in energy production, muscle repair, immune function, and numerous other biological activities (Cruzat et al., 2018). Glutamine is considered conditionally essential because the body cannot always synthesize sufficient amounts to meet its requirements. The need for glutamine increases during periods of physical stress, such as intense or prolonged exercise and when recovering from injury or illness (Coqueiro et al., 2019). When glutamine levels are insufficient, a glutamine-deficient state may lead to immunosuppression and increase the risk of infection (McRae, 2017).

L-glutamine is a popular workout supplement used to counteract the decline in blood glutamine during intense exercise. Studies show that elevating blood glutamine levels can enhance workout recovery by supporting the protein synthesis needed for repairing stressed muscle cells and mitigating inflammatory processes during exercise (Coqueiro et al., 2019; Legault et al., 2015).In a double-blind, placebo-controlled trial, professional basketball players were supplemented with 6 g of L-glutamine per day over 20 days of intense five-hour physical training sessions. The players who took L-glutamine experienced reduced rates of exercise-induced muscle damage, measured as lower blood levels of the enzymes aspartate transaminase, creatine kinase, and myoglobin. Additionally, players who were supplemented with L-glutamine had lower adrenocorticotropic hormone levels, which are produced in response to stress (CórdovaMartínez et al., 2021).

In a randomized, placebo-controlled study, healthy men and women who were supplemented with L-glutamine at a dose of 0.3 g/kg of body weight daily for three days recovered faster from daily knee extension exercises compared to participants who took a placebo. Supplementation was found to accelerate participants’ recovery of muscle force and reduce their muscle soreness at 24, 48, and 72 hours following the eccentric exercises (Legault et al., 2015).

Glutamine depletion contributes to weakened immunity in athletes who overtrain (Cruzat et al., 2018). Because it is essential for proper immune cell function, many athletes supplement with L-glutamine proactively (Cruzat et al., 2018). In a placebo-controlled study, runners were supplemented with approximately 7 g of L-glutamine daily over nine days of twice-daily interval training and five days of rest. Runners taking L-glutamine maintained stronger immunity, measured as 54% higher nasal mucosa immunoglobulin A (IgA) levels (Krieger et al., 2004). Additionally, combat-sport athletes who were supplemented with 0.3 g/kg of body weight for three weeks reduced their incidence of cold symptoms by more than 50% compared to athletes taking a placebo (Lu et al., 2024).

For some individuals, one strenuous workout is enough to disturb the intestinal barrier, triggering inflammation and gastrointestinal distress. Over time, intestinal distress makes the body more vulnerable to infection (Tataka et al., 2022; McRae, 2017). L-glutamine promotes digestive system health during periods of prolonged physical stress by protecting and stabilizing the intestinal barrier (Wang et al., 2018). In a doubleblind, placebo-controlled study, young adults were supplemented with 3 g of glutamine plus 0.69 g of the amino acid cysteine daily for six days prior to engaging in strenuous running exercises. Blood samples collected pre- and post-workout indicated that exercise-induced damage and permeability in the gastrointestinal tract were significantly reduced through glutamine and cysteine supplementation (Tataka et al., 2022).
